UAE TEAM NOMINATES ITS FIRST PRECLINICAL CANDIDATE DRUG

This announcement is made by InSilico Medicine Cayman TopCo (the "Company", together with its subsidiaries, the "Group" or "Insilico Medicine") on a voluntary basis to inform the shareholders and potential investors of the Company about the latest business updates of the Group.

The board of directors of the Company (the "Board") is pleased to announce that Insilico Medicine has officially nominated ISM0387, an MTA-cooperative PRMT5 inhibitor, as a pre-clinical candidate (PCC). Discovered locally by Insilico Medicine's UAE team using the Company's proprietary Pharma.AI platform, the program completed the early discovery workflow – from molecular design to optimization – in the UAE within less than 12 months, during which a series of hit compounds was generated within 30 days, followed by the completion of lead compound optimization within 6 months.

ISM0387 is an MTA-cooperative PRMT5 inhibitor with AI-powered novel structure, showing improved in vitro activity and selectivity, enhanced brain penetrant properties, and robust, dose-dependent efficacy in disease models. Featuring novel structure and exceptional CNS-penetrant abilities, ISM0387 offers a promising new treatment option for patients with Glioblastoma (GBM).

This nomination also represents Insilico Medicine's 30th AI-supported pre-clinical candidate (PCC) to date. The announcement of this achievement was witnessed and supported by the Emirates Drug Establishment (EDE), the Department of Health (DoH) – Abu Dhabi, and the Abu Dhabi Investment Office (ADIO).
