Separately, Phase II Krascendo 170 data presented at ASCO showed that the combination of divarasib with pembrolizumab produced strong efficacy results across PD-L1-positive and -negative cohorts in first-line NSCLC, which management said supported the ongoing Phase III Krascendo 2 trial in the same setting. Roche also disclosed a broader RAS-targeted pipeline including G12D-specific and pan-mutation assets, positioning the franchise for potential use across multiple mutation classes. When analysts asked how divarasib should be evaluated against emerging pan-RAS agents, Graham acknowledged a role for pan-RAS inhibitors but said the high prevalence and specificity of the G12C mutation favored a targeted approach for patients harboring that alteration.

Giredestrant: November PDUFA Approaches Amid Physician Adoption Questions

The PDUFA date for giredestrant in adjuvant estrogen receptor-positive (ER-positive), HER2-negative early breast cancer was set for November 30 under the lidERA trial program, with a second PDUFA for the evERA metastatic program set for December 18. Management said no advisory committee meeting has been indicated to date following priority review.

Analysts pressed on the pace of physician adoption. Graham acknowledged that some KOLs at ASCO raised questions about the absence of longer-term PFS and OS data versus CDK4/6 inhibitors, but said Roche's conversations with potential prescribers indicated broad enthusiasm, driven primarily by tolerability: approximately 50% of patients currently on CDK4/6-based therapy cannot sustain treatment, she said, creating a population of patients either waiting for an alternative or having already discontinued. Management said switch data for patients currently on CDK4/6 therapy is being generated but is not yet available, and that the EU filing is being deliberately delayed to allow the data package to mature.

Schinecker added that giredestrant holds both a potency advantage and a superior safety profile relative to another approved selective estrogen receptor degrader (SERD), without naming the comparator. Analysts from Deutsche Bank and BNP Paribas raised pricing concerns, noting that SERDs have launched above USD 300,000 annually in the metastatic setting, which could constrain broad adjuvant adoption. Graham declined to provide pricing guidance prior to approval but said Roche has historically maintained tighter US-to-ex-US price corridors than some competitors and would price based on clinical value, payer capacity, and the need to sustain R&D investment.

BTK Degrader: Nurix Partnership Adds Targeted Protein Degradation to Hematology and Immunology

Roche closed its collaboration with Nurix Therapeutics to co-develop bexobrutideg (bexdeg), a BTK degrader, during Q2. When asked by Goldman Sachs analyst James Quigley whether the differentiation would manifest primarily in efficacy, safety, tolerability, or combinability, Graham said all four, while acknowledging the asset is early-stage for Roche and that more detail would be provided at Pharma Day.

NXT007 / Zemocimig: Hemophilia Franchise Succession Planning Underway

NXT007 (zemocimig), described by Schinecker as approximately 30 times more potent than emicizumab (Hemlibra), has enrolled its first patient in a Phase III head-to-head trial versus Hemlibra. Bruno Eschli, Head of Investor Relations, said data are expected around year-end 2027 or early 2028. Management said the 360-patient trial design is statistically powered based on known Hemlibra performance and the assumed potency of NXT007.

When Deutsche Bank analyst Emmanuel Papadakis asked whether the trial was designed for superiority or non-inferiority, Graham declined to specify the primary endpoint structure but said the program is intended to demonstrate meaningful patient benefit beyond what Hemlibra currently provides, citing durability and convenience. She characterized antidrug antibodies (ADAs) observed in the Phase I/II study as not clinically meaningful — noting no impact on pharmacokinetics, efficacy, or safety and no cross-reactions — while confirming ongoing monitoring. The Hemlibra full-year growth outlook was revised upward to mid-single-digit from low-single-digit, though management flagged anticipated competitive headwinds in H2 2026 from upcoming competitor launches.

Obesity Pipeline: Multiple Phase III Programs Initiated Across Distinct Mechanisms

The earnings discussion devoted substantial time to the CVRM and obesity franchise. Management confirmed a formal Phase III go-decision for petrelintide monotherapy in obesity, with trial initiation expected in H2 2026. The ZYNERGY combination trial of enicepatide plus long-acting amylin analog petrelintide is expected to initiate in H2 2026, designed to identify the optimal fixed-dose combination for a potential Phase III. Management described the combination rationale as targeting patients requiring either greater weight loss or better glycemic control with improved tolerability.

Enicepatide Phase II data presented at ADA showed that the 24mg dose had not reached a weight loss plateau at 48 weeks, suggesting potential for additional weight loss with longer treatment or higher doses. At the highest dose tested, 48% of patients achieved greater than 20% weight loss, 38% exceeded 25%, and 26% exceeded 30%. Graham said Roche plans to explore doses above 24mg in Phase III. Management declined to identify a single preferred obesity asset, positioning the portfolio as deliberately covering distinct patient segments — deep weight loss (enicepatide), tolerability-driven moderate weight loss (petrelintide), and potential oral dosing (CT-996).

CT-868, a GLP-1/GIP dual agonist previously in development for type 1 diabetes, was discontinued. Management said the decision was unrelated to safety or efficacy and was driven by the view that enicepatide may offer superior potential for glycemic control in type 1 diabetes; detailed plans for that indication were deferred to a later stage.

Analysts from BNP Paribas challenged the differentiation case for both CT-996 and petrelintide. On petrelintide, BNP's Peter Verdult noted that other amylin-targeting compounds have also demonstrated good weight loss and tolerability at lower doses, and asked why no dose response was observed in the Phase II ZUPREME-1 data. Eschli acknowledged that the absence of a dose response had been observed with other gut hormones and said the underlying biology is not fully understood.


Fenebrutinib and Trontinemab: Regulatory Filing Imminent; Long-Term AD Data Presented

Management said the US filing for BTK inhibitor fenebrutinib in MS is expected to be completed "in the coming weeks," with filing acceptance anticipated in Q3 2026. Graham said no signals from FDA interactions suggest the filing will be problematic, and that the FDA allowed open-label extensions for both PPMS and RMS patients — with control-arm participants crossing over to fenebrutinib — which Schinecker characterized as a confidence indicator. The EU filing requires additional data collection and will follow the US package. Investor concerns about liver safety remain live, and management declined to comment on specific regulatory interactions.

Long-term Phase I/II data for amyloid beta-targeted trontinemab in Alzheimer's disease were presented at the Alzheimer's Association International Conference (AAIC) in London the week prior to the call, with management saying the data confirmed rapid and deep amyloid clearance and a strong safety profile. The PrevenTRON Phase III trial, enrolling cognitively unimpaired individuals at high risk of progression to symptomatic Alzheimer's disease, was also disclosed. Management noted the Elecsys pTau217 blood-based Alzheimer's diagnostic — which received CE mark in May 2026 — as a complementary asset.

Gazyva in Immunology: Five FDA Priority Reviews and Multiple H2 PDUFA Dates

Gazyva (obinutuzumab) received FDA breakthrough therapy designation and priority review in membranous nephropathy (MN) with a PDUFA date of November 15, following Phase III MAJESTY data presented at ERA showing superiority over tacrolimus on complete response. US and EU filings in MN have been completed. The INS indication carries a PDUFA date in September 2026, and the lupus nephritis indication is already launched in select markets. Management said patient shares are approaching 10% in markets where lupus nephritis is reimbursed, with early hospital-setting adoption described as strong. The sefaxersen Phase III interim readout in IgA nephropathy is expected in H2 2026 and was described by Schinecker as a potential fourth new molecular entity launch for the year.

Management maintained a peak sales estimate of up to CHF 2 billion for Gazyva across all immunology kidney indications, while acknowledging that indication size, entrenched competitors in SLE, and oncology-anchored pricing constrain the upside case relative to other immunology franchises.
