Abstract
The present invention provides, among other things, improved interleukin-33 (IL-33) variants engineered for increased manufacturability and activity, for example, mutated at one or more of N60, C116, C97, C121 or C148 relative to a truncated IL-33. The present invention also provides a multifunctional fusion protein comprising an IL-33 variant associated with an IL-2 polypeptide and optionally, a half-life extension moiety. The present invention further provides compositions and methods for proliferation and activity of ST2+ regulatory T cells, for prophylaxis and treatment of autoimmune disease.
